Rotenone

What is Rotenone?    

Rotenone is a natural organic compound with the chemical formula C23H22O6 and CAS Registry Number 83-79-4. It is mainly found in the roots of plants in the genus *Rotenone* (Fabaceae family) and the stems, leaves, and seeds of  Dioscorea opposita. It is also distributed in the roots of the traditional Chinese medicinal herbs Dioscorea opposita seeds, Salvia miltiorrhiza seeds, and  Spatholobus suberectus. Its melting point is 159-164℃. It is almost insoluble in water, unstable, and easily decomposes in alkaline substances. When exposed to light and air, it oxidizes, gradually changing the solution from colorless to yellow, orange, and finally deep red, precipitating dehydrorotenone and rotenedione crystals, while still retaining insecticidal activity.


As a selective insecticide, this substance inhibits the activity of NADH dehydrogenase in the mitochondrial respiratory chain of pests, blocking cellular energy metabolism. It has contact and stomach poison effects on aphids, sorghum aphids, and other pests.

Feature

Inhibition of the cellular respiratory chain: Rotenone specifically inhibits mitochondrial complex I (NADH dehydrogenase), blocking electron transport and ATP synthesis, leading to cellular energy depletion and death. This effect is particularly rapid in insects and fish, and the same target is also present in mammalian cells.


Neurotoxicity: Due to the extremely high energy requirements of nerve cells, rotenone can selectively cause damage to dopaminergic neurons. Long-term exposure may induce Parkinson's disease-like symptoms. Its lipid solubility allows it to easily cross the blood-brain barrier, exacerbating the risk of central nervous system damage.

Applications
